摘 要:采用酶法、超声波法、冻融法对金葡球菌破壁,OD54nm值表明:酶法破壁效果最好,超声波法次之。而冻融法有利于金葡球菌细胞结合青霉素酶溶出,超声波法、酶法处理,细胞结合青霉素酶部分失活。0.1%TritonX-100对破壁后细胞结合青霉素酶具有增溶作用。Effects of enzymatic,ultrasonic and freeze-thawing cell ruptures on cell-bound penicillinase activity of Staphylococcus aureus were investigated.OD540nm Values werecompared through the process of cell rupture. It provided evidence that lysostaphin andlysozyme treatments disrupted effectively ceIl walls of S. aureus,ultrasonic treatment wasIess effective,Treatments of cell walls with enzymatic and ultrasonic disruptions not onlyfailed to release any large quantities of cell-bound penicillinase but also might lead to partlyinactivation of it,in contrast,freeze-thaw should be the method of choice for the release ofcell-bound penicillinase.After cell rupture cell-botind penicillinase was soltibilized by 0.1%Triton X-100.
