抗生素89－07与庆大霉素动物体内药代动力学比较研究

摘　　要：分别给狗和大鼠肌肉注射抗生素８９－０７和庆大霉素，作两药在动物体内的药代动力学比较研究，血、尿药浓度采用微生物法测定。结果表明：给狗分别肌注抗生素８９－０７１．５、３．０和４．５ｍｇ／ｋｇ，在０．６０±０．０８、０．６５±０．２３和０．５６±０．２０ｈ达到峰浓度５．３９±０．９９、８．２６±２．１０和１７．４５±２．６８ｍｇ／Ｌ，药时曲线下面积分别为１２．３２±３．４７、２２．７０±４．６８和４１．２３±８．２８ｍｇ·ｈ／Ｌ，消除半衰期为１．０８±０．４３、１．４０±０．２５和１．２０±０．２ｂｈ。肌肉注射庆大霉素３．０ｍｇ／ｋｇ，峰浓度８．３９±２．６０ｍｇ／Ｌ，达峰时间０．７３±０．２１ｈ，药时曲线下面积为２１．９１±７．６３ｍｇ·ｈ／Ｌ，消除半衰期为１．１６±０．２０ｈ．两药具有相似的规律。给大鼠分别肌注抗生素８９－０７和庆大霉素２ｍｇ／ｋｇ，分别于０．２８±０．０９和０．２６±０．１０ｈ达到峰浓度４．８５±１．２３和５．１０±０．６９ｍｇ／Ｌ，消除半衰期为０．５６±０．２３和０．６４±０．１２ｈ，药时曲线下面积为５．７０±２．３４和６．２５±０．６０ｍｇ·ｈ／Ｌ。给狗肌肉注射抗生素８９－０７和庆大霉?Antibiotic 89- 07 was administered intramucularly to dogs and rats,incompare with gentamicin; The concentrations of antibiotic 89-07 and gentamicin in serumand urine were measured by bioassay。 The pharmacokinetics of antibiotic 89-07 weredetermined in dogs after administration of 1.5,3. 0 and 4. 5mg/kg, Mean peak serumconcentrations were reached 5. 39±0. 99,8. 26±2. 10 and 17. 45±2. 68mg/L at 0. 60±0. 08,0. 65±0. 23 and 0. 56±0. 20h respectively。 Mean area under the serum concentration timecurve were 12. 32 ± 3. 47, 22, 70± 4. 68 and 41. 23 ± 8。 28 mg·h/L, respectively, Meanelimination half-life values were 1. 08 ± 0. 43,1. 40 ± 0. 25 and 1. 20±0. 26h. In comparewith gentamicin after administration of 3. 0mg/kg, the phamacokinetics of two drugs weresimilar(Cmax8. 39±2. 60 mg/L,Tpeak 0. 73±0. 21h, AUC 21. 91±7。 63 mg·h/L,T1/2β1. 16±0. 20h); To rats, the Cmax of antibiotic 89-07 and gentamicin following 2mg/kg doses were4. 85±1. 23 and 5. 10±0. 69mg/L,Tpeak were0. 28±0. 99 and 0. 26±0. 10h,AUCwere5. 70±2. 34 and 6. 25±0. 60mg· h/L, T1/2β were 0. 56±0. 23 and 0. 64±0. 12h respectively。Urinary excretion of antibiotic 89-07 and gentamicin after administration of 3mg/kg to dogsaccounted for 85. 11%and 93. 09%of the dose, 76. 76%and 90. 49%to rats. Therefore,the results showed that the pharmacokinetics in animals of antibiotic 89-07 and gentamicinwere similar。
