摘  要:NAU—PAGE(不连续醋酸尿素聚丙烯酰胺凝胶电泳)技术解决了准确、快速测定玉米种子纯度问题。玉米种子贮藏蛋白在杂交种内呈双亲共显性,且主要存在于种胚内。因此,用此方法测定玉米种子纯度不受种子大小、正反交的影响。虫害后只要有胚存在就不影响玉米种子纯度测定。种子萌动后,蛋白质谱带开始模糊,不利于纯度测定。Noncoherent acetic acid urea-polyacrylamide gel electrophoresis(NAU-PAGE) can be used to test maize seed purity precisely and quickly, while clear electrophoretogram can be made under conditions of urea 2.smol/L.stacking gel T=7.5%, C=0.33% , pH5 and running gel T=15%, C=0.67%, PH2.7 with high concentration of acetic acid urea (AL) extracting solution. This technology has solved the important problem of testing maize seed purity quickly and precisely, which demanded prompt solution for a long time. The com seed protein compositions of double parents all emerge in their hybrids and exist mainly in the seed embryo Therefore, the seed purity test would not be influenced by volume, positive and negative crosses and insect damage if only seed maintains emhryo. While com seed begins germinating, the electrophoretograms of seed protein become blurred, which is unfavourable to seed purity test.
