摘  要:应用聚合酶链反应扩增恶性疟原虫环子孢子蛋白质(CSP)基因,扩增产物使用随机引物法进行地高辛标记,所制备的CSP基因探针斑点杂交试验表明,至少可检测出25个单纯虫体,可探及的最低DNA限量为5pg水平。Southern印迹杂交结果分析表明,CSP基因探针可检测出虫体最低DNA水平为6.4pg。In this report, the gene of circumsporozoite protein (CSP) of Plasmodium falciparum were amplified by polymerase chain reaction (PCR) . And PCR products were labeled with digoxigenin by the random primer technique. The result of dot-blotting hybridization assay showed that CSP gene probes could detect at least 25 malaria parasites and 5 pg DNA from Plasmodium falciparum. Also, Southern-blotting hybridization assay indicated that CSP gene probes could detect 6- 4pg DNA. It was evident that CSP gene probes were possessed of a high degree of sensitivity.
