摘  要:应用一株T淋巴母细胞CEM免疫BALB/c小鼠后,取脾细胞与SP2/O职合,分别筛选出3株分泌抗β_2-微球蛋白(β_2-m)单抗细胞株,它们的反应性均能被纯化的可溶性β_2-m完全抑制,但D_(11)E_3、H_5B_2两种单抗既能与游离的β2m反应,亦能与细胞膜表面HLA-β_2-m复合物反应,它们识别相同或者权相近的抗原决定簇。相反,D_8E_3抗体不与细胞膜表面抗原反应,经抗体竞争实验及加成实验,均证实D_8E_3抗体所识别的抗原决定簇,与D_(11)E_3、H5B2不同。推测在细胞膜表面,D_8E_3抗体所识别的抗原决定簇可能被HLA重链相关区域所覆盖。3 cell strains secreting anti-92-microglobulin (β_2-m) McAbs were screened by fusing SP 2/O with the murine splenocytes immunized with T lymphoblast CEM. The reactivities of McAbs secreted by all the 3 strains could be completely inhibited by purified soluble &-m. However, 2 of them (D11E3 and H5B3) could react with either free β_2-m or HLA-5,-m compo.nd located on the surface of cell membrane. It indicated that the antigenic determinants recognized by them were identical or very similar. On the contrary, an other McAb (D11E3) could not react with cell membrane surface antingens, and both antibody competitive and additional tests proved that the antigenic determinants recognized by D8E3 were different from those recogized by D11E3 or H5B2. The authors speculated that the antigenic determinants which could recognize by D8E3 on the surface of cell membrane might be covered by the correlative region of HLA heavy chain.
