摘 要:采用限制性片段长度多态性(RFLP)技术,对90例中国汉族人进行分析,发现载脂蛋白AⅠ(ApoAⅠ)M1等位基因频率在体重指数(BMI)≥24的非胰岛素依赖型糖尿病(NIDDM)与正常对照组之间、在BMI≥24的NIDDM与BMI<24的NIDDM之间、在动脉硬化(AS)与正常对照组之间均存在显著性差异。此外,NIDDM伴AS(NA)的等位基因频率分布与NIDDM相似,与AS不同。根据上述结果,认为:ApoAⅠ基因与BMI≥24的NIDDM相关联;BMI≥24的NIDDM与BMI<24的NIDDM之间存在着遗传异质性;ApoAⅠ基因与AS相关联;在ApoAⅠ基因水平上,未见NIDDM与AS相关联。The restriction fragment length polymorphism (RFLP) technique was used to study 90 Chinese of Han nationality. The subjects were divided into four groups: Non-insulin-dependent Diabetes mellitus (NIDDM),Atherosclerosis (AS),NIDDM with AS (NA) and control subjects (Contronl).Each group was subdivided into BMI≥24 and BMI<24 based on body mass index (BMI). Genomic DNA was digested with the restriction endonuclease Mspl and hybridized with 32p-labelled ApoAⅠ cDNA probe. The results showed that there were significant differences between NIDDM with BMI≥24and Control with BMI≥24,NIDDM with BMI≥24 and NIDDM with BMI<24 and also between AS and control.The allele frequency of NIDDM with AS NA was similar to that of NIDDM but different from that of AS. We believe that there were correlations of ApoAⅠ gene with NIDDM with BMI≥24 as well as AS. There were genetic heterogeneity between NIDDM with BMI≥24 andNIDDM with BMI<24. There were, however, no relationship between NIDDM and AS in ApoAⅠ gene.
