胱胺对离体兔动脉环的舒张作用

摘　　要：用兔离体主动脉环观察腕胺对苯福林缩血管作用的影响以及与一氧化氮代谢的关系１０—１００μｍｏｌ·Ｌ￣（－１）的胱胺在苯福林预收缩的兔主动脉环表现缓慢而完全的舒张作用，呈剂量和时间依赖性，与内皮细胞存在与否无关。１００μｍｏｌ·Ｌ（－１）脱胺预温育５ｈ，使主动脉环对苯福林的收缩敏感性明显降低，ＥＣ＿（５０）增加，最大收缩力下降。胱胺结构类似物半胱胺，半胱氨酸，Ｎ—乙酰半脱氨酸均未见抑制血管收缩力的作用。亚甲基蓝（１０μｍｏｌ·Ｌ￣（－１）），血红蛋白（１μｍｏｌ·Ｌ￣（－１））及Ｎ＿ω—硝基精氨酸（１００μｍｏｌ·Ｌ￣（－１））均可部分反转胱胺对血管收缩力的抑制，使标本的最大收缩力恢复（Ｐ＜０．０５），提高标本对苯福林的敏感性，结果表明，胱胺对血管平滑肌有直接舒张作用，其机理可能与血管平滑肌细胞的一氧化氮代谢途径有关。Cystaminc(CA,10-100μmol·L-1)induced dose-dependent vasorelaxation in rabbitaortic rings(RA R) precontracted with phenylephrine(Phe,1 μmol·L-1).Presence or absence ofendothelium did not influence the vasorelaxation effectof CA. Incubation of RAR with CA(100 μmol·L-1)for 5 h resulted in remarkable reduction of sensitivityto Phe(1 nmol·L-1 -40 μmol·L-1),a right shift ofthe contraciton curve and an increase of EC50.Thedepressant effect was not seen in incubation withcysteamine,cysteine and N-acetyl-cysteine,indicatingthat the depressant action of CA on vascularcontractility was related to structural configurationrather than a specific group present.Haem oglobin(ascavenger of NO, 1 μmol·L-1),methylene blue(aninhibitor of guanylate cyclase,10 μmol·L-1)andN-nitro-L-arginine(an inhibitor of NO-synthase,100 μmol·L-1),all antagonized to some extent thedepressant action of CA on RAR and restored partlythe sensitivity of RAR to Phe,suggesting that effectson the metabolic pathway of NO might be involved inthe vasorelaxation of CA
