摘 要:用跨膜电位及收缩同步记录方法,观察小檗碱(Ber)对豚鼠门静脉平滑肌细胞膜电位及收缩特性的作用。Ber(24.5μmol/L)能引起平滑肌细胞膜静息电位较小的去极化,抑制紧张性(tonic)收缩及自发性收缩频率,增加相性(phasic)收缩幅度。高钾对照组平滑肌细胞膜迅速去极化,紧张性收缩增加,相性收缩消失。Ber能部分对抗K+收缩平滑肌的作用。The effects of berberine(Ber) on transmembrane potential and contraction activity were studied with simultaneous recording technique. The resting membrane potential(RMP) of smooth muscle cells in guinea pig portal vein was -48.8±8.0 mV(n=66).Ber (24.5 Pμmol/L) slightly depolarized the RMP from -47.0±2.0 mV to -44. 6±11. 4 mV (n=11, p<0. 05). The tonic contraction and spontaneous contraction frequencies were decreased. The amplitude of phasic contraction was enhanced. 40 mmol/L K+, as a control group, deplorized the RMP from -51.2±10.3 mV to -42.0±10.5 mV (n=9,p<0. 001), and increased the tonic contraction significantly. The phasic contraction was abolished, and the contractive effect induced by K+ was partly reduced by Ber. These results suggested that Ber could cause vasodilation and membrane depolarization.
