速释硝苯地平微丸的研究

摘　　要：报道了用硝苯地平（Ｎｆ）和聚乙烯吡咯烷酮（ＰＶＰ）制成固体分散体后滚制成Ｎｆ微丸的新工艺。Ｎｆ微丸与市售片剂的溶出度试验表明：其体外累积溶出百分率经０．５ｈ分别为８０％和≤５％，经Ｂｅａｇｌｅ犬口服两剂型的自身对照试验表明，微丸组犬体内血药达峰时明显加快（Ｔ＿（ｍａｘ）＝１ｈ），生物利用度也有显著提高。A new technique was used to prepare nifedipine pellets by spheronization of sohddispersion of nifedipine(Nf),PVP and some other ingredients,The dissolution rates of Nf pelletsand market Nf tablets were 80%and≤5%respectively,Shorter T_(max) and bigger AUC wereproven obviously by pharmacokinetic experiments in Beagles which took the Nf pellets as com-pared with the group of taking the Nf tablets.
