摘 要:目的观察小鼠感染日本血吸虫后早期、急、慢性、晚期肝组织免疫病理与Th1/Th2细胞因子的动态变化.探讨Th1/Th2在肉芽肿及纤维化形成及调节中的作用。方法采用石蜡切片,H E染色后观察感染小鼠不同时期肉芽肿、纤维化病变.用ELISA双抗夹心法测定小鼠感染后4、8、12、20 周脾淋巴细胞IFN-γ和IL-4水平的动态变化。结果小鼠感染后6~8周出现明显肝肉芽肿病变,且肉芽肿反应达高峰,随后下降。感染小鼠脾淋巴细胞的Th1细胞因子IFN-γ在感染早期开始明显上升,急性期升至高水平,随后随感染时间延长呈下降趋势;而Th2细胞因子1L-4在感染后早期亦开始明显上升,且随感染时间延长呈上升趋势。结论 Th1细胞因子IFN-γ、Th2细胞因子IL-4均与血吸虫卵肉芽肿的诱导和形成密切相关,而Th2细胞因子IL-4可抑制Th1细胞因子,在血吸虫病慢性期虫卵肉芽肿与晚期纤维化自发性免疫调节中起重要作用。Objective To observe the dynamics of liver tissue immune pathology in acute,chronic and late stages and the relative changes of Th1 / Th2 cytokine levels in mice infected with Schistosoma Japonicum, and probe the role of Th1/ Th2 on S.Japonicum granuloma and the formation and regulation on fibrosis. Methods Liver granuloma and fibrosis were observed in the different stages for the mice infected with Schistosoma Japonicum by histological examination and the ELASA were used for the quantitative determination of IFN-γand IL-4 in spleen lymphocyte culture medium at 4、8、12 and 20 wk after infection.Results The liver appeared granuloma formation obviously at 6-8 wk and the reaction of granulma reached the peak.Subsequently, it reduced gradually.The Th1 cytokine IFN-γ rose obviously at the early stage and reached to a high level at the acute stage,and decreased gradually with the time of infection.The Th2 cytokine IL-4 also rose obviously at the early stage,but took on an ascendant trend along with the time.Conclusion Both the Th1 cytokine IFN-γ and the Th2 cytokine IL-4 were correlated closely with S.Japonicum granuloma formation and induction in early stage.However,Th2 cytokine IL-4 can inhibit the Th1 cytokine,and it might play an important role in immune regulating on egg granuloma reaction at chronic schistosomiasis and late fibrosis spon-taneously.
