高效液相色谱法同时测定四季青中4种酚酸类成分的含量

摘　　要：目的建立以高效液相色谱法同时测定四季青中原儿茶酸、原儿茶醛、绿原酸、咖啡酸4种酚酸类成分含量的方法。方法 Agilent Zorbax SB-C18柱(4.6mm×250mm,5μm),采用流动相1%冰醋酸(含0.2%三乙胺)-乙腈进行梯度洗脱,流速为1.0ml/min,检测波长280nm,柱温30℃。结果原儿茶酸、原儿茶醛、绿原酸、咖啡酸的线性范围分别为0.480～3.840μg、0.095～0.760μg、0.100～0.800μg和0.110～0.880μg;加样回收率(n=6)分别为99.1%、98.8%、99.3%、99.8%。结论本方法简便、灵敏、准确,具有良好的重复性和稳定性,有利于提高四季青的质量控制水平。Objective To establish a method for simultaneous determination of the content of protocatechuic acid,protocatechuic aldehyde,chlorogenic acid and caffeic acid in Folium Ilicis Purpurea by high performance liquid chromatography (HPLC).Methods An Agilent Zorbax SB-C18 column(4.6 mm×250 mm,5 μm) was used as analytic column with the mobile phase of acetonitrile and 1% acetic acid(including 0.2% triethylamine).Then gradient elution was carried out at the flow rate of 1.0 ml/min.The determination was performed at wavelength of 280 nm and temperature of 30 ℃.Results The linear range of protocatechuic acid,protocatechuic aldehyde,chlorogenic acid and caffeic acid was 0.480-3.840,0.095-0.760,0.100-0.800 and 0.110-0.880 μg respectively;the average recovery (n=6)of these four phenolic acids was 99.1%,98.8%,99.3% and 99.8%,respectively.Conclusion This method is simple,sensitive and accurate with good reproducibility and stabilization,which is helpful to improve the quality control of Folium Ilicis Purpurea.
