摘 要:目的观察二十八烷醇对缺氧大鼠的保护作用,评价其抗缺氧效果,为研发新型抗缺氧药物提供科学依据。方法清洁级雄性成年SD大鼠18只,随机分为平原对照组、缺氧对照组、缺氧给药组,每组各6只。缺氧给药组是在模拟高原环境下(海拔5000m)按5mg/kg剂量给予0.5%(m/v)二十八烷醇悬浊液灌胃,缺氧对照组为相同条件下给予等体积的1ml/kg食用调和油灌胃,平原对照组则在原(海拔300m)直接给予食用调和油灌胃,连续灌胃30d,1次/d。各组大鼠30d后取其心、脑组织于电镜下观察其病理变化。结果缺氧给药组大鼠心、脑组织水肿程度比缺氧对照组减轻,且脑组织比心脏组织效果明显。结论二十八烷醇可提高大鼠的缺氧耐受能力,对缺氧大鼠心脑组织细胞的结构有一定保护作用。Objective To observe the protective effects of 1-Octacosanol on anoxic rats and evaluate its anti-hypoxia effects,and to provide a rationale for the development of a new-type antihypoxic.Methods 18 healthy male SD rats were randomly divided into three groups(n=6):plain control group,hypoxia control group and hypoxia treated group.Hypoxia treated group was exposed to a simulated high altitude(5000 m above sea level) in a low-pressure chamber and intragastrically administered 0.5%(m/v) 1-Octacosanol suspension according to 5 mg/kg dosage;hypoxia control group was intragastrically administered the equal volume of edible oil according to 1 ml/kg dosage under the same condition;plain control group was intragastrically administered edible oil in plain(300 m above sea level).After one lavage per day for 30 d,all the animals were killed rapidly.Their cardiac and cerebral tissues were obtained and the pathological changes were observed under electron microscope.Results Compared with hypoxia control group,the hydropic degree of rat cardiac and cerebral tissues in hypoxia treated group was significantly relieved,especially in cerebral tissues.Conclusion 1-Octacosanol can improve the anti-hypoxia capability of rats and it has certain cytoprotective effects on cerebral and cardiac muscle tissues in hypoxic rats.
