摘  要:目的优化白囊耙齿菌多糖的提取工艺及多糖生物活性研究。方法以超声时间、超声功率和水料比(V/W)为影响因素,应用中心组合试验方法进行实验设计,以多糖得率为响应值,进行响应面分析。利用MTT法检测多糖对大鼠肾系膜细胞增殖的影响。结果多糖提取的最佳条件为:超声时间184 s,超声功率410 W,水料比48∶1;抑制肾系膜细胞的增殖的IC50为16.21μg.ml-1。结论响应面法可用于白囊耙齿菌多糖的提取,其多糖可显著抑制肾系膜细胞的增殖。Objective To optimize the extraction conditions of polysaccharides from Irpex lacteus Fr.and to investigate its bioactivity.Methods Extraction time,ultrasonic power and the solvent/solid ratio were selected as causal factors.Experimental design was developed by central composite design method.The yield of polysaccharides was used as the responsive values,and response surface method was employed to analyze the results of experiments.MTT assay was used to study the effect of polysaccharides on rat glomerular mesangial cells.Results The optimum process conditions for polysaccharides were: extraction time 184 s,ultrasonic power 410 W and the solvent/solid ratio 48:1(V/W).The polysaccharides amount to inhibit the proliferation of glomerular mesangial cell was 16.21 μg·ml-1.Conclusion This method can be applied to the extraction of polysaccharides and the polysaccharides can inhibit the proliferation of rat glomerular mesangial cells.
