摘  要:目的探讨宫内缺氧对40 d龄幼年大鼠齿状回胶质纤维酸性蛋白(GFAP)表达的影响及当归的干预作用。方法配种得孕SD大鼠(39只),至孕14 d,随机分为对照组12只、缺氧组12只、当归组15只。将缺氧组大鼠置于三气培养箱内缺氧,制作宫内缺氧模型,缺氧前1 h经尾静脉注射NS(8 ml/kg),当归组经尾静脉注射当归注射液(8 ml/kg),其余同缺氧组;对照组经尾静脉注射NS,不缺氧;自然生产,产后第40天随机取幼年大鼠脑组织,常规石蜡切片,免疫组织化学染色。结果缺氧组GFAP阳性染色面积(Area)均数比对照组大,差异有统计学意义(P<0.05);当归组Area均数比缺氧组小,差异有统计学意义(P<0.05)。结论 130 ml/L氧浓度的缺氧环境促进了幼年大鼠齿状回GFAP的表达,当归注射液能够减少宫内缺氧后幼年大鼠神经胶质细胞GFAP的表达,对神经系统发挥保护作用。Objective To explore the effects of intrauterin hypoxia on expression of GFAP in dentate gyrus of juvenile rats and the regulative role of angelica on it.Methods Conceiving female SD rats(39) were randomly divided into 3 groups: control group(12),hypoxia group(12) and angelica group(15).The conceiving rats were put into three-gas incubator from the beginning of pregnant 14th day to make intrauterine hypoxia model.In hypoxia group,1 hour before hypoxia,the rats were injected saline from caudal vein;In angelica group,the rats were injected angelica injection,other steps were the same.In control group,the rats were injected saline,no hypoxia.The neoratal rat brains were taken on the 40th day after birth,immobilized,inbeded sliced,and common immunohistochemistry staining were used to detect the Nestin positive substances.Results The average of positive staining area of GFAP in hypoxic group was larger than that of control group,and there had significance(P<0.05).The average of area in angelica group was less than that of hypoxia group,and there had significant difference(P<0.05),Conclusion 1.130 ml/L oxygen environment can promote GFAP expression in dentate gyrus of juvenile rats.2.Angelica injection can reduce GFAP expresion in dentate gyrus of juvenile rats after intrauterin hypoxia,and has protective effect on central neural system.
