摘  要:目的分析人骨髓基质干细胞作为组织工程种子细胞,在长期体外扩增及大规模培养条件下,其染色体核型及端粒酶活性是否发生异常的改变。方法采用常规细胞染色体G显带处理方法及端粒酶活性PCR-ELISA检测法分析3例人骨髓基质干细胞样本。结果人骨髓基质干细胞体外培养至P10代,未发现染色体核型异常改变,相应的端粒酶活性也未出现异常增高。结论人骨髓基质干细胞在体外长期培养扩增的条件下,染色体核型及端粒酶活性未出现异常改变,符合种子细胞安全性的应用要求。Objective To analyze the chromosome karyotype and telomerase activity of human BMSCs which are used as seed cells for tissue engineering during multi-generation culture. Methods Routine methods were used to fix cells and G-banding technique was applied to analyze the chromosome karyotype of human BMSCs. Also Roche's telomerase PCR-ELISA kit was used to detect the telomerase activity of human BMSCs. Result No abnormal changes the chromosome karyotype and telomerase activity of human BMSCs were detected during multi-generation culture till P10 in vitro. Conclusion The chromosome karyotype and telomerase activity of human BMSCs measured up to the security prescript after multi-generation culture.
