摘  要:目的:检测侧支血管生长过程中外膜中成纤维细胞是否活化。方法:左旋支慢性阻塞诱导冠状动脉侧支的生长。采用共聚焦免疫荧光术研究生长中侧支血管外膜中波形蛋白和层粘蛋白(FN)的表达变化;采用电子显微镜观察其成纤维细胞超微结构的改变。结果:生长中侧支血管外膜的成纤维细胞数目剧增,FN的表达明显上调,分别是正常血管外膜中的5倍和8.6倍;生长中侧支血管外膜的成纤维细胞含有大量的粗面内质网和线粒体。结论:在侧支血管生长过程中外膜的成纤维细胞由静止状态变成活跃表型,成纤维细胞的激活对侧支血管的外向生长可能具有重要作用。To test whether adventital fibroblasts are activated during collateral vessel growth or not. Methods: Coronary collateral vessel growth was induced by chronic occlusion of the left circumflex coronary artery. Expression of vimentin and fibronectin in the adventitia was detected using confocal immunoflurescence. In addition, ultrastructure of fibroblast was examined by transmission electron microscope (TEM). Results: The number of fibroblasts was significantly increased in the adventita in growing collateral vessels, being 5 times higher than that in normal vessels. Expression of fibronectin was significantly increased in growing collateral vessels, being 8.6 times higher than that in normal vessels. TEM revealed that growing collateral vessels fibroblasts contained numerous rough endoplasmic reticula and mitochondria. Conclusion: Fibroblasts in the adventitia are activated during the growth collateral vessel. The expression of fibronectin is increased, indicating that activated fibroblasts may play an important role in the outgrowth of collateral vessels development.
