摘  要:采用放射免疫检测方法,观察了强啡肽(Dyn)和苯环利定(PCP)对高血压大鼠(SHR)的环腺苷酸(cAMP)生成系统的影响.结果表明:Dyn剂量依赖地降低SHR和非高血压大鼠(WKy)的外周血管中cAMP的含量,并且对SHR的cAMP生成系统的抑制作用大于WKy(P<0.05);相反地,PCP剂量依赖地升高SNR和WKy的外周血管中的cAMP的含量,对SHR的cAMP生成系统的激活作用小于WKy(P<0.05)。这些观察表明:1.Dyn和PCP通过抑制或激活环腺苷酸生成系统对SHR产生作用。2.SHR和WKy的外周动脉对Dyn和PCP的敏感性不同。P<0.01vsWKy.EffatOf ̄oncAMPconalsof ̄neP ̄Of ̄ ̄inSHRandWKyIn ̄ngconrmhationofPCP(10--'--10--'mol/L)induceadoSedrpendentinCreaseincAMPleVelsinmembranepa ̄ofbothSHRandWKy.TheEC.valueSwere9.3x10--7mol/LforWKyand5.0x10--6mol/LforSHR.The effect of the kappa-receptor agonist dynorphine (Dyn) and the phencyclidine (PCP) -receptor agonist PCP on 3' -5' -cyclic adenosine monophosphate (cAMP)generating system of the spontaneously hypertensive rat (SHR) arteries was studied in vitro. Dyn decreased cAMP levels in a dose-dependent manner in both SHR and normotensive Wistar Kyoto rat (WKy) peripheral arteries, but the inhibitory effect of Dyn was statistically greater in the SHR arteries than that in the WKy ones. PCP increased cAMP levels in a dose-dependent manner in both SHR and WKy peripheral arteries. The stimulatory effect of PCP was statistically greater in the WKy arteries than that in the SHR ones. The findings indicate that (1) Dyn and PCP can affect SHR and WKy arteries through inhibition or activation of the adenylate-cyclase cAMP system, (2) the sensitivity of SHR arteries to Dyn and PCP was different from that of WKy arteries.
