摘  要:大量证据表明,兴奋性氨基酸(excitatoryaminoacids,EAAs)是脊椎动物中枢神经系统的兴奋性神经递质,并参与伤害性信息的一级传递。但对NMDA受体在不同伤害性信息传递中的作用却颇有争议。本实验采用免疫组化方法显示脊髓Fos样免疫活性(Fos—likeimmunoreactivity,FLI)神经元,观察化学性伤害性刺激物福尔马林(5%,150μl)足底注射对脊髓c—Fos表达的诱导,以及MK—801对福尔马林诱导脊髓Fos蛋白产生的影响。结果:福尔马林足底注射引起的Fos表达主要分布于脊髓背角的Ⅰ—Ⅱ层和Ⅴ-Ⅵ层;MK—801(0.6mg/kg,i.p.)几乎完全阻断福尔马林引起的脊髓Fos表达。这一结果表明,福尔马林刺激所引起的脊髓Fos蛋白表达,绝大多数由NMDA受体介导,提示NMDA受体在福尔马林化学性伤害性信息传递中,具有至关重要的作用。Excitatory amino acids (EAAs) have been shown to be a sort of very important excitatory neurotransmitters in the central nervous system of vertebrates and involved in the primary signal transduction of noxious stimuli. However there are controversies about the role of NMDA receptors in the transduction of different kinds of noxious stimuli. In the present study, we use immunocytochemical staining of c-Fos protein to demonstrate the effect of intraplantar injected formalin (5%, 150μl) on the Fos induction in rat spinal cord, as well as the effect of MK-801, a non-competitive NMDA antagonist, on the formalin-induced Fos expression. The results were as follows: (a) formalini-nduced Fos -like immunoreactivity (FLI) neurons mainly distributed in lamina Ⅰ-Ⅱ and lamina Ⅴ-Ⅵ of the spinal gray matter; (b) formalin -induced FLI neurons were almost thoroughly blocked by pre-administration of MK-801 (0.6 mg/kg, i. p.). The results indicate that formalin -induced Fos expression in the spinal cord is mainly mediated by NMDA receptors, suggesting the key role of NMDA receptors in the signal transduction of formalin chemical noxious stimuli.
