摘  要:本文用大鼠进行实验,观察了长时间电针刺激时脑脊液和中脑导水管周围灰质内CCR-8样免疫活性物质(CCK-8-ir)含量,并用Northern印迹法测定脑内CCKmRNA含量的变化。结果表明,电针1h后大鼠脑脊液中CCK-8-ir的浓度增高,2h后中脑导水管周围灰质的含量也明显升高,8h后大鼠脑内CCK-8mRNA含量显著增加。表明长时间电针加速脑内CCK—8的生成和释放,最终加速了CCK基因表达,这可能是CCK-8参与电针耐受的机理之一。Experiments were performed in rats in order to observe the effects of prolonged electroacupuncture (EA) stimulation on the content of immunoreactive (ir) cholecystokinin octapeptide (CCK-8) in CSF and in the tissue of periaqueductal gray (PAG).Changes in the brain content of mRNA coding for preprocholecystokinin was also determined by the Northern blot technique. The results showed a significant increase in the content of ir -CCK-8 both in the spinal perfusates and in the tissue of PAG at the end of 1h and 2h of continuous EA stimulation, respectively.The brain content of CCK mRNA did not show any significant increase until 8h of continuous stimulation. It is concluded that prolonged EA stimulation elicited an accelerated release of CCK-8 and the processing of CCK -8 from its precursors, followed by a delayed response of CCK gene expression. This may constitute one of the mechanisms for the development of EA tolerance.
