摘 要:许旺细胞是周围神经系统的髓鞘形成细胞.它能修复遗传性或物理、化学因素导致的中枢脱髓鞘损伤、本实验用荧光素Hoechst33342标记体外培养的人胎儿许旺细胞并将其移植于1%溶血卵磷酯造成的脱髓鞘大鼠脊髓内,直接观察许旺细胞的成活和迁移。实验结果显示人胎儿许旺细胞特异性地朝脱髓鞘损伤方向迁移,迁移的速度约2mm/d,迁移途径主要是软脊膜。室管膜以及血管周间隙.亦有部分细胞在白质内迁移、移植1个月后,细胞保持迁移能力,荧光素标记的细胞核轮郭清楚;两个月后,细胞迁移能力明显减弱,细胞稀疏结果表明,人胎儿许旺细胞在大鼠脊髓内可成活2个月个并对脱髓鞘损伤信号具有反应性。Human fatal Schwann cells labeled with the nuclear fluorochrome Hoechst 33342 in vitro migrated at a distance of 6 mm in the rat spinal cord with lysolecithin us induced lesion.Our result showed that some transplanted human fetal Schwann cells migrated toward the lesion site in the rat spinal cord at an average speed of 2 mm/d. The migration of transplanted Schwann cells was preferentially along the ependyma, makinged and blood vessels as well as white matter tracts. Four weeks after the transplantation, Schwann cells could still keep their ability of migration, labeled nucleus of Schwann cells manifesting clear profile under fluorescence microscopy, although the number of Schwann cells decreased & weeks after transplantation. This suggests that cultured human fetal Schwann cells could survive at least for & weeks in the spinal cord of rat and were able to respond to stimulation of demyelinated lesion.
