摘 要:基于从东亚钳蝎中鉴定的低电导Ca2+激活K+通道毒素BmP05的蛋白质和前体核苷酸序列,采用5′和3′RACE(rapidamplificationofcDNAends)技术,成功地从东亚钳蝎毒腺组织中分离到了一系列与BmP05前体核苷酸序列高同源性的cDNA序列.它们含有完整开放阅读框(openreadingframe,ORF),彼此同源性很高,有的突变导致编码不同氨基酸,有的只是沉默突变.基因组克隆结果进一步显示,尽管来源不同个体蝎的BmP05基因组的结构相同,但它们的基因组序列并不完全一致,在第二个外显子和内含子区域发生许多突变.BmP05在cDNA水平的多态性与其在基因组水平的多态性一致.所有结果表明,东亚钳蝎的毒素基因存在个体多态性.Based the amino acid sequence and the on the precursor nucleotide sequence of small conductance Ca^2+-acctivateel K^+ channel (SKCa) toxin gene BrnP05, a series of cDNA sequences were isola- ted and identified from Chinese scorpion venom of Buthus martensii Karsch, using the 3' and 5' RACE (Rapid amplification of cDNA Ends) methods. Their precursor nucleotides all contained a complete open reading frame(ORF). These cDNA sequences also shared high homology with each other: some mutations resulted in the variance of amino acid residue and the other mutations did not result in the changement of amino acid. Genomic cloning of BmP05 was shown that although the genomic organization and structure of BrnP05 from different scorpion individuals were identical with each other, their genomic sequences were not completely same, whose mutations took place in region of the intron and the second exon. The genomic polymorphism of BmP05 toxin gene was consistent with the cDNA polymorphism at the RNA level. All these results showed that individual polymorphism of toxin genes from Buthus martensii Karsch took place.
