摘 要:目的:通过不同的中药炮制方式,降低大黄的遗传毒性.方法:采用不同中药炮制方式炮制生大黄,应用鼠伤寒沙门菌体外回复突变试验(Ames)和彗星实验(Comet assay)对生大黄及不同炮制方式炮制后的大黄的遗传毒性进行研究.结果:生大黄Ames试验中TA97、TA102菌株无论代谢活化或非代谢活化皆为阳性;而除醋蒸样品在代谢非活化条件下呈阳性外,各菌株的清蒸和醋蒸大黄Ames试验均为阴性,但清炒和醋炒大黄仍为阳性.彗星实验结果显示,生大黄在1.25,2.5和5.0mg/mL浓度下,其尾长、尾部DNA含量与阴性对照组相比差异有统计学意义(P<0.01),且其毒性呈剂量-效应关系,大黄各剂量组清蒸和醋蒸与阴性对照组比较差异无统计学意义,达到了降低毒性的效果.而清炒和醋炒的样品仍具有遗传毒性.结论:生大黄具有一定的遗传毒性,清蒸和醋蒸两种炮制方式可以有效的降低大黄的遗传毒性,而清炒、醋炒减毒效果不明显.Objective:To reduce the genotoxicity of rheum officinale by different processing method.Method:After processing rheum officinale with different methods,the Ames test and Comet assay were used to detect the genotoxicities of the processed rheum officinales and non-processed rheum officinale.Result:Reverse mutation were induced on TA97 and TA102 after treated with rheum officinale no matter whether there is S9 metabolism activation or not.Rheum officinale after steamed with or without vinegar all showed negative results except TA102 after treated with steamed rheum officinale with vinegar without S9 metabolism activation.The results of Comet assay showed that,there was no significant differences(P>0.05) of rheum officinale steamed with or without vinegar Compared with the negative control group,and there were significant differences(P<0.01) of raw rheum officinale.Further more,it shows a dose-dependent effect of 1.25,2.5 and 5.0mg/mL rheum officinale with the tail length and the tail DNA%.Compared with the negative control group,there was no significant differences(P>0.05) of rheum officinale steamed with or without vinegar,which showed great reduction of rheum officinale genotoxicity.There was still genotoxicity of rheum officinale after stir-frying with or without vinegar.Conclution: rheum officinale could cause genotoxicity,while rheum officinale steamed with or without vinegar could largely reduce genotoxicity.Rheum officinale stir-fried with or without vinegar could still cause genotoxicity.
