摘 要:目的研究神经干细胞小神经球体外和脊髓内移植后的分化。方法从20周人胚胎分离神经干细胞进行鉴定,经培养成小神经球体外诱导分化;成年雌性Wistar大鼠于T11水平行椎板切开术并横切腰段脊髓,立体定向注射细胞50000个进入两侧脊髓断端中线的3个部位(薄束、楔束及皮质脊髓束)。移植3个月后利用双标免疫荧光观察神经干细胞的分化情况。结果小神经球实验组在体外和受伤大鼠脊髓内诱导分化的神经元高于单细胞悬液对照组,前者有显著差异,后者有极显著差异。小神经球脊髓移植后细胞的存活率明显增加,与对照组相比有显著差异(P<0.05)。结论小神经球移植增加细胞的存活率,分化的神经元数量增加,应当成为较好的脊髓神经干细胞移植方式。Objective To identify small neurospheres differentiation potential in vitro and in rat spinal cord. Methods Isolate and identificate hNSCs(human neural stem cells) from human fetus of about 20 weeks. Small neurospheres in culture were induced to differentiate. Adult Wister rats were performed a sterile laminectomy at Tll and the spinal cords were transected. 5 × 10^4 hNSCs were sterotaxicly injected into three sites of the midline of both cord stumps(ventral funiculus, dorsal corticospinal tract,gracile fasculus) immedialy after transection. Three months after transplantation, cells' survival and differentiation were tested with immunohistochemistry. Results Small neurospheres' (test groops) generated a higher proportion of neurons in vitro and in injured rat spinal cords than single cells (control group) ;the fomer had significant difference( P 〈 0.05), the latter had very significant difference( P〈 0.01 ). Cells survival rate of small neurospheres graft groups increased significantly than control groups( P 〈 0.05). Conclusion Small hNSCs neuropheres transplantation may provide a better spinal cord transplantation means for increase number of cell survival and neuronal differentiation.
