摘 要:目的通过观察人参皂苷(GS)诱导人T淋巴细胞白血病细胞株(Jurkat细胞)凋亡,探讨GS影响T淋巴白血病细胞增生作用的机制。方法选用人T淋巴细胞株(Jurkat细胞)作靶细胞,采用MTT法和半固体集落培养法观察不同浓度的GS对Jurkat细胞增生的抑制效应;用An-nexinV-FITC试验法分析Jurkat细胞的凋亡百分率,并进行DNA片段分析(DNALadder)。结果MTT法和半固体集落培养法均显示GS在50μg/mL浓度下能够明显抑制Jurkat细胞增生,并随浓度增加抑制作用增强;流式细胞术显示GS在一定浓度和时间范围内可引起细胞凋亡。结论GS在一定浓度范围内可抑制Jurkat细胞增生,并能够诱导其凋亡,为临床应用提供理论依据。Objective To investigate whether ginsenosides(GS) could induce Jurkat cells apoptosis to explore the mechanism of GS in T lymphocyte leukemia. Methods The inhibition effects of GS on Jurkat cell line were observed by using proliferation test of MTT and semi-solid colony formation. The apoptosis rate and DNA ladder were analyzed by Annexin-V FITC test, DNA Electrophoresis respectively. Resuits GS (50μg/mL) could inhibit the proliferation of Jurkat cells with significant difference between GS and non-GS control (P〈0.05) in both MTT test and colony assay. And the inhibition effects were increased by the concentration of GS. GS could induce Jurkat cells apoptosis in a certain scope of concentration and reacting time. Conclusion GS could inhibit the growth of Jurkat cells and induce them apoptosis, which provides an experimental basis for treatment of lymphocyte leukemia.
