摘 要:比较分析了4种可以应用于光学和电子显微观察的方法:透明压片法、石蜡切片法、半薄切片法以及超薄切片法.结果表明,上述4种方法均可用于鉴定银杉外生菌根,但对于外生菌根的早期发育阶段,透明压片法比其他方法简单方便;对于外生菌根完整结构的观察宜采用半薄切片法.Four available methods for light or electron-microscopic observation were analyzed: clearing and staining method, paraffin section method, semi-section method and ultra-thin section method, The results indicated that all these methods were able to identify ectomycorrhizal roots of C, argyrophylla. Especially, an early developmental stage of ectomycorrhizal roots identified by the clearing and staining method is easier than other methods. Alto intact structure of ectomycorrhizal roots were observed only by semi-section method.
