摘 要:目的探讨生长激素(Growth hormone, GH)对大鼠阿霉素(Adriamycin,ADR)心肌病心肌细胞凋亡及其相关基因蛋白BCL-2、BAX表达的影响.方法 30只雄性Wistar大鼠,体重200~250 g,随机分为3组:对照组、ADR组和GH+ADR组.用原位末端标记法(TUNEL)标记凋亡的心肌细胞,用免疫组织化学方法检测BAX和BCL- 2的蛋白表达.结果与对照组比较,ADR组心肌细胞凋亡指数明显升高(P<0.05),GH+ADR组细胞凋亡指数低于ADR组(P<0.05).ADR组BAX蛋白表达明显高于对照组(P<0.05), BCL- 2蛋白表达明显低于对照组(P<0.05).GH+ADR组BAX的蛋白表达明显低于ADR组(P<0.05),但高于对照组(P<0.05);BCL- 2的蛋白表达明显高于ADR组(P<0.05),接近对照组.结论心肌细胞凋亡是导致阿霉素心肌病的主要原因, GH干预治疗可减少阿霉素心肌病的心肌细胞凋亡,这可能与GH能提高BCL- 2/BAX比值有关.Objective To explore the effects of growth hormone (GH) on cardiomyocyte apoptosis and the expression of BCL-2, BAX protein in adriamycin (ADR) -induced cardiomyopathy in rats. Methods Thirty male Wistar rats weighing 200 - 250 g were randomized into three groups : control group, GH + ADR group and ADR group. Apoptotic cardiomyocytes were detected using the terminal deoxynucleotidyl transferase mediated dUTP nick end labeling method (TUNEL). The expression of BAX and BCL-2 proteins was determined by immunohistochemical method. Results Compared with control group, ADR group had significantly higher index of apoptotic cardiomyocytes (P 〈 0.05). The apoptotic index in GH + ADR group was less than that in ADR group (P 〈 0.05). The expression of BAX protein in ADR group was significantly higher than that in control group ( P 〈 0.05), while the expression of BCL-2 protein was significantly lower than that in control group ( P 〈 0.05). The expression of BAX protein in GH + ADR group was significantly lower than that in ADR group ( P 〈 0.05 ), but significantly higher than that in control group ( P 〈 0.05). The expression of BCL-2 protein in GH + ADR group was significantly higher than that in ADR group ( P 〈 0.05 ), and was closed to that of control group. Conclusions Myocardial apoptosis is an important reason of adriamycininduced cardiomyopathy. GH therapy can inhibit eardiomyocyte apoptosis in adriamycin-induced cardiomyopathy, partly because it might increase the ratio of BCL-2 /BAX.
