摘 要:目的探讨β2肾上腺素受体(β2AR)基因多态性与支气管哮喘发病及其严重程度的相关性。方法采用扩增阻滞突变系统聚合酶链反应(ARMS-PCR)技术来检测β2AR基因16和27位点基因型在45例哮喘患者与他们的健康一级亲属(对照组)中的频率分布。结果(1)哮喘组16位点Gly/Gly基因型频率为31%,健康对照组中频率为11%,两者比较差异有显著性(P<0.01);而27位点两组各基因型频率分布无显著性差异(P>0.05);(2)轻度哮喘患者16位点Gly/Gly基因型频率为9.5%,中重度哮喘患者频率为37.5%,两者比较差异有显著性(P<0.01),而27位点两组各基因型频率分布无显著性差异(P>0.05)。结论β2肾上腺素受体(β2AR)基因多态性与哮喘发病及其严重程度之间有一定的关系,可能是哮喘的易感基因之一。Objective To investigate the correlation between the genetic polymorphisms of β2- adrenergic receptor gene and the development and severity of asthma. Methods The 16 and 27 polymorphisms of β2-adrenergic receptor gene was genotyped by amplification refractory mutation system-polymerase chain reaction(ARMS-PCR) in 45 asthmatic subjects and their healthy direct relatives. Results (1) The frequency of Gly/Gly genotype in 16 site was 31% in asthmatic group and 11% in control group with a significant difference between two groups(P〈0.01); there was no significant difference in the distributions of the genotypes in 27 site between two groups(P〉0.05). (2) The frequency of Gly/Gly genotype in 16 site was 9.5% in the patients with mild asthma and 37.5% in the patients with moderate severe asthma (P〈0.01); there was no significant difference in the distributions of the genotypes in 27 site of two groups(P〉0.05). Conclusion There is a certain correlation between the genetic polymorphisms of β2-adrenergic receptor gene and the development and severity of asthma;β2-adrenergic receptor gene may be one of susceptibility genes.
