摘  要:目的研究正常衰老小鼠小脑颗粒细胞膜膜电位及诱发动作电位的变化情况。方法老年(20~24月龄)和青年(3~4月龄)C57/BL6小鼠,以振荡切片法制备小脑脑切片作为观测对象,运用全细胞记录的电生理方法测定脑片上小脑颗粒细胞膜膜电位和诱发动作电位,并进行比较。结果在无任何刺激的静息条件下,衰老小鼠的小脑颗粒细胞能够维持其细胞膜电位的稳定;对于衰老神经元,诱导产生动作电位所需的内向电流降低,同时动作电位的时程延长。结论衰老虽然不影响神经元在静息条件下的状态,但影响神经元的动作电位以及其后的恢复程度:衰老神经元更容易丧失其膜电位。Objecetive To study the alterations in neuronal membrane potential (MP) and the induced action potential (AP) in cerebellar granule neurons in brain sections of aged mice. Methods Cerebellar sections (200μm thickness) were made from young (3-4 months old) and old (20 - 24 months old) C57BL/6 mice, and the whole - cell recording was used to detect the MP and the induced AP of granule neurons. Results The granule neurons of aged mice could manage to maintain the MP values in resting condition, while the input - current needed to evoke a AP decreased and the duration of AP increased remarkably in an Ⅰ - clamp study. Conclusions Although aging did not show an influence on resting MP, it did change the AP and its recovery. Aged neurons were more vnlnerable and tended to lose their MP after recording.
