摘 要:文章针对ATP生产的现场检测过程,建立了一个以离子交换树脂为分离介质的色谱分析方法,并对其分析效果进行了试验研究。色谱条件为:D201树脂柱(Φ6 mm×500mm),流动相分别为pH值3.0的乙酸溶液和pH值9.0、1.5 mol/L的NaCl溶液,检测波长254 nm,流速10 ml/min,室温23±2℃。检测重复性较好, 在0.1-8 g/L范围内,ATP浓度与ATP峰面积呈较好线性关系,r=0.997 7;平均回收率为98.60%.整个色谱过程可在30min内完成。In the process of ATP production, a kind of chromatography method has been set up to analyze ATP and AMP. The separating medium is macroporous anion-exchange resin D201, and the effects of the analytic procedure have been discussed through experimental investigations. The chromatographic conditions were selected as follows: D201 resin column(φ6mm×500mm); the mobile phase was acetic acid solution (pH3. 0) and NaC1 solution (pH9.0,1.5mol/L) ; detected at 254nm; flow rate was 10 ml/ min, and column temperature was 23±2℃. The method has good repeatability and good linearity within the range of 0.148 g/L(r=0. 9977) between CATF and ATP peak area. The average recovery was 98. 60%. The process can be completed within 30 min.
