摘 要:目的:研究黄芪注射液对体外培养的神经细胞的缺血、缺氧损伤的保护作用。方法:在体外培养大鼠胎鼠的大脑皮层细胞,通过去除培养液中的血清造成细胞损伤,实验组加入不同浓度的黄芪,观察黄芪对上述损伤的作用;用氰化钠造成神经细胞的缺氧模型,比较黄芪治疗组和对照组的乳酸脱氢酶(LDH)、K+的流出量的变化,同时以MTT法测定存活细胞数。结果:缺血24h后,黄芪治疗组的线粒体活性值明显高于对照组,且有一定的量效关系;对于缺氧损伤的细胞,黄芪治疗组的LDH、K+流出量明显低于对照组,且细胞存活数明显高于对照组。结论:黄芪具有一定的抗神经细胞缺血缺氧损伤的作用。Objective: To investigate the protective effects of Astragalus against anoxic and ischemic lesion on the neural cells in vitro. Methods: Brain cortical cells of fetus rats were culturing in vitro, and bovine serum was removed from culture media in order to induce cells apoptosis with different concentration of Astragale added into serum groups, the influence of Astragale on the neural cells were observed; The hypoxic damage mode of neural cells were induced with NaCN, and one group was treated with Astragale , then a the LDH and K + effluxes in both groups were compared, the survival cells count was determined with MTT. Results: After taking off bovine serum, the cell mitochondrial activity treated with Astragale group was significantly higher than that of non - Astragalus; the effiuxes of LDH and K ^+ were increased after 24 hours of anoxic , hut the effluxes of the group treated with Astragalus was much more lower than that of non - astragalus and the survival cells count was much high than that of non - astragalus as well. Conclusion: Astragalus can protect the cultured rat neural cells against anoxic damages in the vitro.
