产甘油假丝酵母产甘油关键酶基因的克隆、测序及植物表达载体的构建

摘　　要：用PCR的方法,从产甘油假丝酵母WL2002—5中扩增出了产甘油关键酶3-磷酸甘油脱氢酶(glycerol3-phosphate dehydrogenase)和3-磷酸甘油磷酸酶(glycerol3-phosphate phosphatase)的基因GPD和GPP。对这两个基因测序,发现基与酿酒酵母(Saccharomyces.cerevisiae)的GPD、GPP基因的相似性达99%。并构建了其在植物中的表达载体。Two genes of GPD and GPP encoding glycerol 3 - phosphate dehydrogenase and glycerol 3 - phosphate phosphatase, two key enzymes in glycerol synthesis, were cloned from Candida glycerol＇genesis WL2002 -5 using PCR （polymerase chain reaction）. After sequencing the cloned GPD and GPP it was found the identity of the two genes with the corresponding genes in Saccharomyces cerevisiae was up to 99%. Furthermore, in order to study their functions in some plants, the plant expression vectors were constructed successfully.
