摘  要:目的探讨缺血预处理对心肌细胞凋亡及血浆TNF水平的影响。方法选择大鼠随机分为假手术组、持续缺血组、缺血再灌注组和缺血预处理组,以活结结扎左冠状动脉的前降支,分别造成阻断冠脉血流和再灌注。并通过以缺口末端标记法(TUNEL)标记凋亡细胞,采用放射免疫法等检测血浆TNF水平等指标判定其结果。结果凋亡细胞原位标记与半定量分析表明:缺血预处理组与持续缺血组、缺血再灌注组比较心肌细胞凋亡程度明显降低(P<0.01)。而缺血预处理组与缺血再灌注组比较,降低TNF的作用很明显,差异十分显著(P<0.01)。结论缺血预处理能降低TNF水平,对抗TNF引起的心肌损害,减轻心肌细胞凋亡程度,保护心肌细胞。Objective To study the effect of ischemic preconditioning on the apoptosis and the TNF plasma level. Methods Wistar rats were randomly divided into sham - operated group, ischemia reperfusion group and IPC group . Ligate the anterior descending branches of the left coronary artery with slipknot to block the blood flow and reperfuse the myocardium. The apoptotic cell was labeled by TUNEL, the level of TNF was determined by RIA. Results In situ apeptotic cell labeling and semiquantitative analysis reveal that the level of apoptosis decreased significantly in the IPC group compared with that in continuous ischemia group and ischemia reperfusion group ( P 〈 0.001). The level of TNF decreased significandy in IPC group compared with that in ischemia reperfusion group, the difference was significant. Conclusion IPC can decrease the level of TNF, resist the myocardial injury resulting from TNF, lessen the degree of apoptosis and protect the myocardial cells.
