摘 要:目的:观察复方丹参(SM)和甲基强的松龙(MP)联合治疗对急性脊髓损伤大鼠后肢运动功能及脊髓前角细胞凋亡和诱导型一氧化氮合酶表达的影响。方法:用30只2个月龄Wistar大鼠随机均分为甲基强的松龙(MP)治疗组(A)、MP+SM治疗组(B);用改良Allen's法致伤脊髓,3周后对大鼠进行斜板试验、行为学评分,HE染色和TUNEL染色及免疫组织化学染色。结果:斜板试验和行为学评分以及脊髓前角内NOS阳性细胞率和细胞凋亡指数,A、B两组比较差异有显著性统计学意义。结论:SM联合MP治疗可以进一步促进急性脊髓损伤大鼠后肢运动功能的恢复,减少前角细胞凋亡;其机制可能与复方丹参提高大鼠损伤脊髓前角内NOS表达有关。Objective To investigate expression of inducible nitric oxide synthase(iNOS) and the apoptosis in anterior horn of spinal cord and evaluate hind limb motor function of rats with spinal cord acute injury(SCI) after treated with salvia milhiorrhiza(SM) and methylprednisolne(MP) . Methods Thirty Wistar rats (2 months) were divided randomly into MP group(group A) and MP + SM group (group B). Spinal cords were injured by Allen's methods of weight drop. Rats'behavior was recorded by BBB score 3 weeks later after injury. All rats were tested by oblique plane, then the cord T10 segment were collected and stained by methods of hematoxylin and eosin(HE),immunohistochemistry and TUNEL staining. Results There were significant different in BBB score (5.30 ± 1.24 vs 7.23 ± 1.91, P 〈 0. 05), oblique board test(32.87±3.11 vs 39.07±5.05, P 〈 0.01), TUNEL positive cells(14.34±2.51 vs 11.29±1.80, P 〈 0.01) and positive neuron of iNOS expression(9.45 ± 1.25 vs 12.69±2.03, P 〈 0.05) in group B and group A. The inclined plane technique showed the joint application group recovered better. The number of NOS positive neurons of NOS in group B was more than the group A. The apoptosis index in group B were less than the group A. Conclusion It is demonstrated that MP combined with SM could promote the recovery of bilateral hind limbs of the rats and prohibit cells apoptosis after SCI spinal cord injury. The reason may be SM promote enhance synthesis of NO in the anterior horn of spinal cord of SCI rats.
