运输应激猪组织病理性损伤、HSP_(27)的组织分布及其相关性研究

摘　　要：利用组织学、免疫组织化学以及临床血液检测等方法,探讨持续运输应激对育肥出栏猪组织的病理性损伤以及组织损伤与HSP27组织定位之间的相关性。结果表明:运输应激初期(1～2h),反映组织损伤的血液肌酸激酶(CK)和天门冬氨酸氨基转移酶(AST)活性都显著升高;应激4h时,CK及AST活性有所下降;当应激持续到6h和10h时,CK活性又显著升高。组织病理损伤也呈现出相似的波动变化。免疫组织化学结果显示,随着应激时间的延长,HSP27在组织中的定位没有发生肉眼可见的变化;HSP27主要分布在心肌和肝细胞胞浆中,在背最长肌的细胞浆和细胞核中均呈强阳性表达,在肺脏细支气管的黏膜下层有强阳性表达,在受检组织的血管壁及内皮细胞中也有强阳性表达。Relationship between tissue damages and heat shock protein 27 （ HSP27 ） distribution in transport stressed pigs after 1 h, 2 h, 4 h, 6 h and 10 h long distance transportation was studied by means of blood parameter detection, histopathological and immunohistochemistric methods. Significant increase of the serum creatine kinase （CK） and the aspartate aminotransferase （AST） which refer to myocardium and hepatocyte damages were occurred at the beginning of transport stress （1 -2 h）. After a slight reductions for 4 h transportation, the serum CK of the transport stressed pigs increased obviously for 6 h and 10 h transportation. With the extension of the transport stress time, the same trend between pathological lesion and CK in serun was found. No significant difference of HSP27 distribution in the detected tissues could be found between the transport stressed and untransport stressed pigs. HSP27 was mainly distributed in the cytoplasm of the myocardium and the hepatocyte. The positive staining signals were distributed in the cytoplasm and the nucleus of musculus longissimus dorsi. HSP27 staining were also detected obviously in the submucosa of bronchiole, blood vessel wall and endothelial cells of all detected tissues.
