摘  要:目的探讨银杏叶提取物对糖尿病大鼠肺泡巨噬细胞功能及诱导型一氧化氮合酶(iNOS)、CD36与过氧化物酶体增生物激活受体γ(PPAR-γ)基因的影响。方法测定各组大鼠血清葡萄糖、胰岛素含量,检测肺泡巨噬细胞超氧化物歧化酶、一氧化氮合酶活性,丙二醛、一氧化氮含量,用RT-PCR测定iNOS、CD36与PPAR-γ基因表达水平。结果①银杏叶提取物对糖尿病大鼠有降血糖作用(P<0.05),并使血清胰岛素水平呈一定程度的升高;②经银杏叶提取物治疗后糖尿病大鼠肺泡巨噬细胞超氧化物歧化酶活性、PPAR-γmRNA表达升高(P<0.05),CD36mRNA表达增加(无显著差异),iNOSmRNA表达、一氧化氮合酶活性、丙二醛和一氧化氮含量均下降(P<0.05)。结论推测银杏叶提取物对糖尿病大鼠的治疗作用可能与刺激胰岛β细胞分泌胰岛素,增强巨噬细胞抗脂质过氧化作用,以及促进PPAR-γmRNA的表达有关。Objective To study the effects of ginkgo biloba extmct(GBE) on the function of alveolar macrophages (AMS) from diabetic rats. Methods Serum glucose and serum insulin concentmtion in different groups were measured. The activities of superoxide dismutase (SOD) and nitric oxide synthase (NOS),as well as the contents of malondialdehyde (MDA) and nitric oxide (NO) in AMs were assayed. In addition, PPAR-γ, iNOS, and CD36 gene expression levels were measured by RT-PCR. Results ① GBE could increase the level of serum insulin, but decrease the level of serum glucose of diabetic rats (P〈0.05). ②The activity of SOD (P〈0.05) and the expression of PPAR-γ gene (P〈0.05) were increased, but the activity of NOS (P〈0.05), the contents of MDA (P〈0.05) and NO (P〈0.05 ), as well as the expression of iNOS gene (P〈0.05) were decreased in GBE-treated group compared with the diabetic group. Conclusion GBE could improve the function of macrophage by adjusting PPAR-γ gene expression and reducing lipid peroxidation in diabetic state.
