摘  要:以5、10、50、100μmol/L维生素C(vitC)及其与10ng/mlTNF-α联合处理体外培养的大鼠脂肪细胞24h,采用流式细胞仪检测脂肪细胞凋亡率,光学显微镜和透射电镜观察脂肪细胞形态学变化。结果表明,高浓度vitC(100μmol/L)单独作用及与TNF-α联用,均能显著提高脂肪细胞凋亡率(P<0.01),且联用组凋亡率显著高于单用TNF-α处理组(P<0.01);低浓度vitC(5、10、50μmol/L)单独作用未检测到凋亡,且与TNF-α联用组凋亡率显著低于单用TNF-α处理组(P<0.01);光学显微镜和透射电镜观察可见100μmol/LvitC处理组出现凋亡的形态学特征,细胞收缩变圆,染色质致密浓染聚集在核膜边缘,核碎裂成多个凋亡小体,随即被邻近细胞吞噬。结果提示高浓度vitC(100μmol/L)可以引起脂肪细胞凋亡,并有效地增强TNF-α对脂肪细胞的凋亡作用。Primary rat adipocyts were treated with 5, 10, 50, 100μmol/L vitamin C (vit C) and combination of vit C (5-100μmol/L) and 10 ng/ml TNF-α, respectively, to investigate their effect on adipocyte apoptosis. The apoptotic rate of adipocytes was measured by flow cytometry. The morphologic change of fat cells was observed with optical microscope and transmission electron microscope. The results show that both higher concentration of vit C (100 μmol/L) alone and its combination with TNF-α could significantly increase apoptosis rate of adipocytes (P〈0.01) and the effect of combination with TNF-α was more apparent than TNF-α alone (P〈0.01). No apoptosis was detected in adipocytes treated with lower dose of vit C (5-50 μmol/L) and the TNF-α-induced apoptosis was markedly inhibited by these concentrations of vit C. The feature of apoptosis was observed in adipocytes treated with 100μmol/L vit C, i.e. cellular shrinkage and round, condensation of chromatin adjacent to nuclear membrane, nuclear fragmentation of apoptotic bodies which were rapidly engulfed by neighboring cells. The data suggest that high dose vit C (100μmol/L) may cause adipocyte apoptosis and markedly enhance TNF-α-induced apoptosis.
