摘 要:[目的]探讨煤工尘肺发生发展过程中血清含Cu抗氧化酶CuZn-SOD和铜蓝蛋白(CP)的活性及Cu含量的变化。[方法]分别采用亚硝酸盐形成法、对苯二胺盐酸盐法及原子吸收分光光度法,测定70例不同期别煤工尘肺患者(Ⅰ、Ⅱ、Ⅲ期)及30例健康对照者的血清CuZn-SOD活性、铜蓝蛋白(CP)活性及Cu的含量。[结果]对照组、Ⅰ期尘肺组、Ⅱ期尘肺组及Ⅲ期尘肺组的血清CuZn-SOD活性分别为(15.96±2.26)、(17.24±2.28)、(19.81±3.01)、(21.63±3.52) NU/ml;血清CP活性分别为(2.26±0.52)、(2.68±0.67)、(2.97±0.79)、(3.36±0.88)U/ml;血清Cu含量分别为(12.68 ±3.87)、(14.96±4.42)、(17.84±5.12)、(19.38±5.89)μmol/L。与对照组相比,各期别尘肺患者血清与CuZn-SOD和CP 活性及其Cu的含量均显著升高。[结论]煤工尘肺的发生发展涉及含Cu抗氧化酶活性的上调,且其活性的改变与Cu含量变化有平行关系。[ Objective ] To explore the changes of serum copper-containing antioxidant enzymes activities including CuZn-SOD, ceruloplasmin ( CP ) and Cu contents in the pathogenesis of coal wocker's pneumoconiosis ( CWP ). [ Methods ] The activities of serum CuZn-SOD, CP and Cu contents in 70 patients with different stage of ( CWP ) ( Ⅰ , Ⅱ , Ⅲ ) and 30 healthy subjects were systematically determined with nitrite forming method, p-phenylene diamine hydrochloride method and atomic absorption spectrophotoassay ( AAS ), respectively. [ Results ]The activities of serum CuZn-SOD in the control group, Ⅰ stage CWP group,Ⅱ stage CWP group and Ⅲ stage CWP group were ( 15.96 ± 2.26 ), ( 17.24±2.28 ), ( 19.81± 3.01 ), ( 21.63±3.52 ) NU/ml, respectively, the activities of CP were ( 2.26±0.52 ), ( 2.68±0.67 ), ( 2.97±0.79 ), ( 3.36±0.88 ) U/ml, respectively, and the contents of Cu were ( 12.68 ± 3.87 ), ( 14.96 ± 4.42 ), ( 17.84 ± 5.12 ), ( 19.38 ± 5.89 )μmol/L, respectively. As compared with the control subjects, the activities of serum CP, CuZn-SOD and Cu levels in patients with CWP of different stages significantly increased. [ Conclusion ] The pathogenesis of CWP might be related to the up-regulation of activities of copper-containing enzymes including CP, CuZn-SOD and the changes of these enzymes activities are also closely correlated with the changes of Cu contents.
