摘 要:目的探讨针刺疗法对阿尔茨海默病(Alzheimer’sdisease,AD)的治疗作用和机制。方法18只快速老化的SAMP8小鼠随机分为针刺组、非穴组、SAMP8对照组(P8组),每组6只,6只正常老化的SAMR1小鼠作为SAMR1对照组(R1组)。针刺组采用“益气调血,扶本培元”针法;非穴组针刺非穴点;P8及R1对照组捉抓,均每日1次,共14日。处理结束后,制备脑切片,作甲苯胺蓝染色,光镜下观察,并用病理图像分析系统进行定量分析。结果P8组海马和颞叶皮质神经元形态发生退行性改变,尼氏体平均总面积和积分光度、平均单位面积细胞数目、平均单个细胞面积及平均CA1区厚度均明显小于R1组(P<0.01);针刺组神经元形态接近R1组,上述指标均大于P8组(P<0.05),平均单位面积细胞数目在CA3区与R1组无差异(P>0.05),在颞叶皮质大于R1组(P<0.05),尼氏体平均总面积和积分光度、平均单个细胞截面积及平均CA1区厚度均小于R1组(P<0.01)。结论“益气调血,扶本培元”针法可以改善SAMP8小鼠神经元形态和功能状态,减少或延迟神经元的丢失,对阿尔茨海默病可能有较好的治疗作用。Objective To investigate the therapeutic action of acupuncture on Alzheimer' s disease (AD) and its mechanism. Method Eighteen SAMP8 were randomly divided into acupuncture, non-point and SAMP8 control (P8) groups, 6 mice each. Six normal senile mice, namely R1 anti-senile accelerated mice made up a SAMR1 control group ( group R1 ). The acupuncture group was treated by qi-replenishing, blood-regulating, kidney-reinforcing and constitution-strengthening acupuncture, the non-point group by non-point acupuncture, and P8 and R1 control groups by gripping. They were treated once daily for 14 days. Brain sections were made at the end of treatment, stained with toluidine blue and examined under an optical microscope. A quantitative analysis was made by a pathologic image analysis system. Results In group PS, hippocampus and temporal cortex neurons had a retrograde change in morphology, and the average total area and integral luminosity of the Nissl body, the number of average unit area cells, the average section area of single cell and the average thickness of area CA1 were all significantly smaller than in group R1 (P 〈0.01 ). In the acupuncture group, the neurons were near in morphology to those in group R1, the above indices were all significantly larger than in group P8 (P 〈0.05 ), the number of average unit area cells in area CA3 was not significantly different from that in group R1 (P 〉 0.05 ) , that in the temporal cortex was significantly larger than in group R1 (P 〈0.05 ), and the average total area and integral luminosity of the Nissl body, the average section area of single cell and the average thickness of area CA1 were all significantly smaller than in group R1 (P 〈 0.01 ). Conclusion Qi-replenishing, blood-regulating, kidney-reinforcing and constitution-strengthening acupuncture can improve the morphology and function of SAMP8 neurons and reduce or delay the loss of neurons and may have a curative effect on Alzheimer' s disease.
