匹多莫德对免疫功能缺陷小鼠脾组织TNF-α和IL-6表达及相关免疫药理作用的研究

摘　　要：目的:研究匹多莫德注射剂对正常小鼠及免疫功能低下小鼠脾组织TNF-α和IL-6表达及相关免疫功能的影响,以及匹多莫德免疫调节作用可能的机制。方法:RT-PCR法对小鼠脾组织的TNF-α和IL-6的表达进行分析;中性红法对小鼠的腹腔巨噬细胞的吞噬功能进行研究;MTT法检测ConA和LPS分别刺激的小鼠脾淋巴细胞的体外增殖;血清溶血素法检测小鼠血清抗体的产生。结果:匹多莫德注射剂连续给药14d对正常小鼠及免疫功能低下小鼠脾组织的TNF-α和IL-6的表达有一定的促进作用;小鼠腹腔巨噬细胞的吞噬能力明显提高;ConA或LPS刺激的小鼠脾淋巴细胞的转化能力均有明显的提高。结论:匹多莫德可明显提高正常小鼠和免疫功能低下小鼠的免疫功能,并促进TNF-α和IL-6表达。AIM：To study the influence of injection agent of pidotimod on function of the immune-system in normal and immune-depressed mice.and to investigate the meehamisms of the immunomodulating action of pidotimod.METHODS：the expressions of TNF-α and IL-6 in mice spleen was detected by BT-PCR method.The effects of pidotimod on phagocytosis functions of mice peritoneal exudate cells was investigated by nentral red phagoeytosis.The lymphocyte proliferations induced by Con A or LPS were detected by MTT method.Adopt the serum haemolysis to measure the production of sernm antibody.RESULTS：The normal and immune-depressed mice were treated for 14 days with different dosages of pidotimod by injection.Pidotimod can significantly inerease the expressions of TNF-α and IL-6 of spleen,potentiate phagocytosis of the peritoneal exudate cells,potentiate the lymphocyte proliferations ability induced by Con A or LPS.CONCLUSION：Pidotimod can potentiate amelioration normal and immune-depressed miee immunesystem function and increase expressions of TNF-α and IL-6 of spleen.
