摘  要:目的:观察逆灸对随后佐剂性关节炎大鼠早期和继发期下丘脑中枢核转录因子P65的影响。方法:实验于2003-09/11在北京中医药大学针灸学院针灸机理实验室进行。将36只Wistar大鼠分为6组,每组6只:①正常组:不造模,不预灸。②模型早期组:大鼠右后足掌皮下注射福氏完全佐剂0.1mL造成佐剂性关节炎模型,造模第3天取材。③逆灸早期组:于造模前进行艾灸,大椎穴处施灸,每次1壮,隔日灸1次,共灸8次,于灸结束后造模,造模第3天取材。④模型继发组:造模第16天取材。⑤逆灸继发组:于造模前进行艾灸,于灸结束后造模,造模后第16天取材。⑥逆灸正常组:正常鼠艾灸不造模,灸结束后取材。下丘脑核转录因子P65阳性细胞的表达强度以吸光度代表,应用免疫组织化学的方法和计算机图像分析。进行组间对比分析。结果:36只大鼠均进入结果分析。①与正常组比,模型各组下丘脑中核转录因子P65阳性颗粒染色明显增强,早期强于继发期(97.60±7.04,359.96±34.93,258.43±33.11,P<0.01)。逆灸早期核转录因子P65阳性染色明显减弱,继发期则接近正常组(242.44±34.61,191.77±10.42)。②逆灸虽可使正常大鼠下丘脑核转录因子P65阳性表达有升高的趋势,但未出现统计学意义(104.32±12.74,P>0.05)。结论:佐剂性关节炎大鼠下丘脑核转录因子阳性细胞处于高表达状态,使细胞因子超量生成,细胞损伤。逆灸可使随后佐剂性关节炎大鼠早期和继发期下丘脑核转录因子P65高表达状态明显减弱,从而对随后佐剂性关节炎的免疫性炎症产生保护作用。AIM: To explore the effect of preventive moxibustion on the hypothalamus transcription factor P65 at the early and secondary stage in rats with succedent adjuvant arthritis. METHODS: The experiment was conducted in Acupuncture and Moxibustion Mechanism Laboratory of Acupuncture School of Beijing University of Chinese Medicine from September to November 2003. Thirtysix Wistar rats were divided into 6 groups with 6 rats in each group: ① Normal group: They were not built into model or acupuncture pretreatment. ② Early model group: Right hindfoot palm was injected with 0.1 mL Freund complete adjuvant to make adjuvant arthritis model to draw the materials at the third day after building model, ③ Early stage preventive moxibustion group: Moxibustion was conducted before building model, and at Tachui (Du 14) moxibustion was done with one moxa-cone at every time and once every two days for eight times. After the end of moxibustion model was made, and materials were drawn at the third day for building model. ④ Secondary model group: Materials were drawn at the 16^th day for building model. ⑤ Sccondary reverse acupuncture group: Moxibustion was done before building the model and models were built at the end of moxibustion. Materials were drawn at the 16^th day for building model. ⑥ Normal reverse acupuncture group: The moxibustion of normal rats did not build model, and materials were drawn at the end of moxibustion. The expression intensity of hypothalamus nucleus transcription factor P65 positive cells was represented by absorbance with immunohistochemical method and computer image analysis in order to compare among those groups. RESULTS: Totally 36 rats were involved in the result analysis. ① By comparison with the rats in normal group, the positive staining of hypothalamus transcription factor P65 significantly increased in every model group, and it was strengthened in the early stage compared with the secondary stage(97.60±7.04, 359.96±34.93, 258.43±33.11, P 〈 0.01 ).
