摘 要:目的探讨即刻早期基因c fos在甲基汞神经毒性分子机制中的作用。方法应用RT PCR方法研究大鼠注射甲基汞急性暴露后,中枢神经系统c fosmRNA表达。结果甲基汞在剂量为0.05mg/(kg·d)暴露60min和剂量为0.5mg/(kg·d)暴露20min就能够显著诱导大鼠脑即刻早期基因c fos表达;c fosmRNA表达量与汞的浓度没有明显正比关系。结论甲基汞能够显著诱导大鼠脑c fosmRNA表达,c fosmRNA表达变化在甲基汞神经毒性分子机制中具有重要作用,即刻早期基因c fos参与了甲基汞对中枢神经系统损害的毒性过程。Objective To study the role that immediate early expression gene(IEG) c-fos played in the neurotoxic mechanism, and the effect of methylmarcury(MMC) on c-fos mRNA expression were investigated. Methods RT-PCR was used for c-fos mRNA expression after MMC acute exposure in rat' s brain. The control group was physiological saline of 0.9%, the concentration of exposure groups were 0.05, 0.5 and 5 mg/kg respectively, the sampling time were 20, 60,240 and 1440 min. Results The 0.05 mg/kg group induced the expression of c-fos obviously in rat's brain after exposed for 60min while the 0.5 mg/kg group induced the expression of c-fos clearly in rat's brain after exposed for 20min, and the expressive quanti- ty of c-fos mRNA did not have direct ratio with the concentration of mercury. Conclusion The expression of c-fos mRNA played an important part in the neurotoxic mechanism of MMC; IEG c-fos took part in the process that MMC impaired the nerve system.
