摘 要:目的:研究青蒿琥酯体外诱导U937细胞凋亡及其过程中细胞内游离钙的变化。方法:采用光镜、透射电镜技术、DNA片段化电泳、流式细胞术对青蒿琥酯诱导U937细胞凋亡进行了观察,并应用激光共聚焦显微镜技术检测凋亡过程中细胞内游离钙浓度的变化。结果:6.250mg/L的青蒿琥酯可诱导U937细胞凋亡,作用于细胞后30min,Ca2+浓度即明显升高,1h达高峰,48h仍维持在较高水平。结论:青蒿琥酯可诱导U937细胞凋亡,细胞胞质内Ca2+是青蒿琥酯诱导U937细胞凋亡信号传导过程中重要的信号分子。Objective:To study apoptosis and the intracellular free calcium of U937 cell induced by artesunate in vitro. Method:Cell apoptosis was investigated by light microscope, ultrastructural changes, DNA gel electro phoresis and flow cytometry. The intracellular free calcium were detected by laser scanning confocal microscopy during the apoptotic process in U937 cells. Result: Artesunate induced U937 cell apoptosis at 6. 250μg/ml. In U937 cells treated with artesunate for 30 minutes, the concentration of intracellular free calcium increased greatly, reached to peak at 1h, and remained the higher level for 48 h. Conclusion: Artesunate may induce apoptosis of U937 cell in vitro. Intracellular free calcium is an important signal molecule in the process of signal transduction of apoptosis in U937 cells.
