摘 要:目的 研究氟桂利嗪、拉莫三嗪及两药联合经孕鼠给药,对胎鼠寓内缺血缺氧性脑损害的保护作用。方法 将孕20天wistar大鼠40只,随机分为A组(氟桂利嗪组,n=10),B组(拉莫三嗪组,n=10),C组(氟棒利嗪+拉莫三嗪组,n=10)和D组(对照组,n=10)。前三组于动物脑缺血缺氧术前3天开始胃管给药,每天1次,剂量分别为氟桂利嗪0.5mg(kg·d),拉莫三嗪10mg(kg·d),氟桂利嗪0.5mg(kg·d)+拉莫三嗪10mg(kg·d)。第二次给药后3小时,每组取8只孕鼠制作宫内缺血缺氧模型,取2只孕鼠不作缺血缺氧处理,剖宫产取胎鼠,每胎5只。于缺血缺氧后6小时、12小时、24小时、48小时分别取胎鼠血清0.5ml,采用ELISA法分别测定血清NSE、S-100水平。结果 孕鼠缺血缺氧后,胎鼠血清NSE和S-100水平较不缺血的明屁升高,氟桂利嗪组、拉莫乏嗪组及两药合用组与对照组缺血缺氧后胎鼠各个时问点血清NSE和S-100水平相比较均降低,差异有显著性意义(P〈0.01)。联合用药较单个药物应用效果更明湿,组间比较差异有显著性意义(P〈0.01)。结论 产前预防性口服氟棒利嗪、拉莫三嗪对官内缺血缺氧性脑损伤具有保护作用,联合用药效果更明显。Objective To investigate the neuroprotective effects of antenatal oral flunarizine,Lmotrigine and the combination of two drugs on changes of neuro specific enloase and S - 100 protein in fetal rats after hypoxic ischemic brain damage. Methods 40 cases pregnant wistar rats(pregnancy for 20 days) were randomly assigned into flunarizine group( n= 10), lamotrigion gronp(n= 10), flunarizine + lamotrigion group(n= 10)and control group(n= 10). Drugs were administered through feeding tube 3 days before hypoxic-ischemic operation. The dosages for the first three groups respectively are: flunarizine0. 5mg( kg· d), lamotrigion10 mg( kg· d), flunarizine0. 5 mg( kg· d) + lamotrigion10 mg(kg·d). Three hours after the third dose,8 cases of pregnant rats from each group were operated for fetal rats hypoxic- ischemic brain damage model,2 cases for control. 5 fetal rats were taken out by caesarean birth from each pregnant rat. Blood samples were taken from fetal rats 6h, 12h,24h,48h after the birth. The concentration of neuro speeific enolase and S- 100 protein in .serum were detected by enzyme immunoassay method. Results The concentrations of serum NSE and S- 100 protein of hypoxic - ischemic fetal rats were higher that that of no ischemic group. The concentration of serum NSE and S - 100 protein in flunarizine group, larnotrigion group, flunarizine + lamotrigion group were lower than that of control group, there were significant difference between groups and between corn bined drugs group with single drug group( P〈 0.01 ). Conclusions Preventive antenatal oral flunarizine has positive neuroprotective effects on intrauterine hypoxie- ischemic brain damage.
