摘  要:采用超声技术提取中药枇杷叶及其药渣中的乌索酸和齐墩果酸,并建立了HPLC!PAD测定乌索酸和齐墩果酸含量的方法。色谱柱为Nova!PakC18(3.9mm×300mm,5μm),流动相甲醇-水(88∶12),二极管阵列检测器检测,检测波长210nm,柱温25℃。乌索酸线性范围:2.28~15.96μg,线性回归系数0.99999;齐墩果酸线性范围:1.18~8.26μg,线性回归系数0.99997。样品中乌索酸的平均回收率为99.6%,RSD为1.3%;齐墩果酸的平均回收率为98.5%,RSD为1.9%(n=5)。结果表明枇杷叶药渣中乌索酸和齐墩果酸的含量均高于枇杷叶原料,用HPLC法测定乌索酸和齐墩果酸含量操作简便,结果准确,线性范围宽,可用于评价含有乌索酸和齐墩果酸药材的质量。The ursolic acid and oleanolic acid from Eriobotryajaponica (Thunb.) LindL and the residues of decoction were extracted with the ultrasonic technology. A determination method of ursolic acid and oleanolic acid was established with the high performance liquid chromatograph and photodiode array detector. The chromatographic column, Nova-Pak C18(3.9×300 mm, 4 μm), methanol-water (88:12, V/V) mobile phase with 0.8 mL/min flow rate, the detected wavelength (210 nm), and the column temperature (25 ℃) were adopted. The calibration curve ofursolic acid was linear under the content of 2.28-15.96 μg and the correlation coefficient were 0.999 99. The calibration curve of oleanolic acid was linear under the content of 1.18-8.26 μg and the correlation coefficient were over 0.999 97. The average recovery of ursolic acid was 99.6 % and RSD was 1.3 %(n=5). The average recovery of oleanolic acid was 98.5 % and RSD was 1.9 %(n=5). The result showed that the content of ursolic acid and oleanolic acid in the residues in the decoction was higher than in the material. The method was simple, accurate and had wider linear range. The method can be adapted in the evaluation of the quality of medicinal plants that contained ursolic acid and oleanolic acid.
