摘  要:朊病毒病(又称传染性海绵状脑病)的病原是致病型朊蛋白(PrPSc),聚集在牛脑中能够引发牛海绵状脑病(BSE)。利用自制的单抗4C11和国外同类单抗进行免疫组织化学(IHC)的比较染色,结果显示单抗4C11和国际上常用的单抗在BSE阳性切片的IHC染色结果上没有显著差异;利用单抗4C11结合IHC方法检测中国山东省150例牛脑样品的实验表明,所有送检样品中IHC的检测结果均为阴性,而且IHC方法比较适合于我国BSE的检测;对中枢神经组织内细胞型PrPC的IHC定位结果显示,PrPC在牛脑组织不同部位的分布有差异。A converted form of the normal cellular prion protein (PrP^c) accumulating in the bovine brains induces bovine spongiform encephalopathy (BSE). This so-called scrapie-associated prion protein (PrP^Sc) found in many animals and humans is the pathogen ofprion diseases. Using monoclonal antibody 4C11 (which was raised against PrP mature protein of Chinese yellow cattle) and the other two kinds of monoclonal antibodies, 6H4 and F89/160. 1.5, which were internationally used, no obvious immunohistochemistic differences were detected in all bovine brain sections of 150 samples from Shandong Province. The result showed that all bovine brain samples were completely negative. With its operating conveniently, diagnosing easily and costing lowly, immnohistochemical (IHC) staining is favorite to detect BSE. The result also showed that the distribution ofPrPc in the different parts of the central nervous system (CNS) tissues from BSE negative brains were different.
