摘 要:目的切除山羊双侧卵巢(OVX山羊)建立绝经后骨质疏松(PMO)动物模型,应用骨组织形态计量学方法观察OVX山羊对四环素-雌酮(XW630)的治疗反应。方法31只雌性山羊随机分成4组,即正常对照组、假手术组、OVX术后6个月组、XW630治疗6个月组,制备山羊髂骨不脱钙骨切片,应用骨组织形态计量学方法,观察各组髂骨骨计量学参数的变化。结果正常对照组和假手术组比较各参数间差异无显著性。与正常对照组和假手术组比较,OVX术后6个月组的骨小梁体积比全部骨组织体积(TBV/TTV)、骨小梁体积比海绵骨体积(TBV/SBV)、平均骨小梁板厚度(MTPT)、四环素双标线距离(DDL)、平均类骨质宽度(MOSW)、骨矿化沉积率(MiAR)和组织水平的骨形成速率(Svf)显著减少(Svf:P<0.05,其余各参数:P<0.01),骨小梁表面比体积(S/V)则显著增加(P<0.01),说明去卵巢后山羊骨量丢失,骨小梁体积明显减小,骨小梁厚度下降、连接性降低,骨转换率降低,激活频率减慢,骨形成明显减少,表现为低转化型骨质疏松的骨代谢特征。骨质疏松山羊模型复制成功。与OVX术后6个月组相比,XW630治疗6个月组的TBV/TTV,TBV/SBV,MTPT,DDL,MOSW,MiAR和Svf显著增加(P<0.01),S/V,骨矿化延迟时间(MLT)显著减少(P<0.01);与假手术组相比,XW630治疗6个月组的TBV/TTV,TBV/SBV显著减少(P<0.01)。结论XW630有增加骨量,改善骨小梁空间结构,增加骨强度,提高骨激活频率,促进类骨质生成及矿化,增强成骨细胞活性的作用。Objective To evaluate effects of XW630 by bone histomorphometry on postmenopausal (PMO) goats models who were established by ovariectomy. Methods Thirty -one female goats were randomly divided into four groups. Group C was for goats without any treatment and Group S was for sham - operated without any treatment. Group OVX6M was for those whose body weight 30kg or below and XW 630 was administered 87.5mg per week while to those whose body weight more than 30kg was 105mg per week. Tetracycline was given to the goats on the 21st and 9th days prior to sacrifice in order to label the bone for dynamic study. Ilium for each goats were reserved for further bone histomorphometric study. Results Trabecular Bone Volume/Total Tissue Volume (TBV/TTV), Trabecular Bone Volume/Sponge Bone Volume (TBV/SBV), Mean Trabecular Plate Thickness( MTPT), Distance between the Double Lables (DDL) ,Mean Osteoid Width (MOSW), Mineral Appositional Rate (MiAR)and Svfin OVX6M group were showed to be significantly lower than those in other groups (P 〈 0.01 ). The data suggest that there were significant increasing in TBV/TTV ,TBV/SBV ,MTPT, DDL,MOSW MiAR and Svf (P 〈 0.01 ) while decreased in S/Vand MLT (P 〈 0. 01 ). In XW6M group the OVX goats models had been successfully established and comparing with OVX 6M Group the XW630 might reverse the process of bone loss and accelerate bone turnover characterized by the increase in activation frequency. Conclusions XW630 seemed to be a promising new medicine to treat PMO by reducing bone loss and increasing trabecular plate thickness and connectivity accelerating bone activation frequency and promoting bone formation.
