金牡蛎胶囊免疫化学鉴别及含量测定的研究

摘　　要：采用多种电泳和免疫方法，确证牡蛎蛋白中，分子量５５ＫＤ等组分为该种属特征蛋白（抗原）。以此组分制备免疫纯对照品，及以其抗血清制备专属检测试药。在奥氏免疫扩散法中，不与其他７种常见贝类交叉。在定量免疫电泳中，标准曲线Ｙ＝３．５２Ｘ－１．１３．ｒ＝０．９９３．加样回收率＝１０３％，ＲＳＤ％＝２．８（ｎ＝１０），测定金牡蛎胶囊含该种属蛋白１７．８±１．２２ｍｇ／丸。初步表明本法用于牡蛎制剂内在质量比较及控制，符合专属，灵敏，快速及简便的要求。By using SDS - PAGE, Western blot and ELISA methods,a 55KD protein was found tobe the specific protein(antigen) of the oyster.With the help of this immunologically pure control and spe-cific antibody,the capsule and other 7 mollusks were identified by Ouchterlony’s immunodiffusion,andquantitative determination of the protein in oyster extract was performed by immunoelectrophoresis rock-et,the rate of recovery being 103% and the relative standard deviation being 2. 8(n=10). It was shownthat the methods are specific and sensitive with good reproducibility.
