摘  要:目的:探讨愈消通脉散对糖尿病大鼠脑组织超微结构的影响及其作用机理。方法:采用链脲佐菌素左下腹腔注射所致糖尿病大鼠模型,采用HE染色、原位杂交、透射电镜等方法,分别检测愈消通脉散对糖尿病大鼠脑组织超微结构及海马ET-1mRNA、ncNOSmRNA、iNOSmRNA、ecNOSmRNA表达的影响。结果:模型组大鼠海马神经元出现退行性变及毛细血管损伤;中药治疗组大鼠海马神经元及毛细血管未见明显病理改变。中药组大鼠海马ET-1mRNA阳性表达明显弱于同期模型组(P<0·01);中药组和对照组大鼠海马ecNOSmRNA阳性表达较模型组明显增强(P<0·01)。结论:持续高血糖状态可以引起糖尿病大鼠海马神经元退行性变及毛细血管损伤,愈消通脉散具有防治糖尿病大鼠海马神经元退行性改变及血管损伤的作用,其机理可能与其具有调节糖尿病大鼠脑组织ET-1mRNA、ncNOSmRNA和ecNOSmRNA表达水平有关。Purpose:To investigate the effects of yuxiaotongmaisan on the encephalic supermicrostructure of rats in diabetes mellitus and its mechanism. Method: the model of diabetes mellitus was established by injection of streptozotocin into the abdomen sinister inferior of rats. H&E staining, in situ hybridization and transmission electron microscope were used to assess the effects of yuxiaotongmaisan on the encephalic supermicrostructure and the expression of ET-1 mRNA, ncNOSmRNA, iNOSmRNA, eeNOSmRNA in hippocampuses of rats in diabetes mellitus. Result : Neuron degeneration and capillary damage appeared in the rats of model group, whereas no obvious pathological changes were detected in yuxiaotongmaisan treatment group. In the corresponding period, the positive expression of ET-1 mRNA in hippocampus of the rats in yuxiaotongmaisan treatment group is apparently weaker compared to the model group( P〈0.01 ). In addition, the possitive expression of ecNOSmRNA in hippocampus of the rats in both yuxiaotongmaisan treatment group and control group seemed to be evidently stronger compared with the model group. Significant as the difference existed in both the amount of positive cells and the mean light density (P〈0.01). Conclusion: yuxiaotongmaisan has preventive and therapeutic effects on neuron degeneration in hippocampus and capilary damage induced by persistent diabetes mellitus, the potential mechanism may relate to the regulation of the expression of ET-1 mRNA,ncNOSmRNA,ecNOSmRNA in hippocampuses of rats in diabetes mellitus.
