类风湿关节炎患者自身反应性T细胞受体氨基酸序列研究

摘　　要：目的研究类风湿关节炎(RA)患者体内自身反应性T细胞的抗原互补区3(CDR3)的序列特性。方法采用实时定量聚合酶链反应(PCR)的方法扩增带有BV14和BV16的自身反应性T细胞抗原受体β链,用基因扫描的方法分析抗原互补区3的片断长度,用序列测定的方法分析CDR3的氨基酸序列。结果RA患者关节滑膜浸润T细胞呈寡克隆扩增,带有BV14和BV16的自身反应性T细胞分别取用不同VDJ片段组合,自身反应性T细胞CDR3长度有15bp、21bp和24bp三种形式,在不同患者自身反应性T细胞CDR3中发现相同氨基酸序列。结论RA患者自身反应性T细胞在病灶部位受到某些抗原的驱动而发生受体取用的偏移和CDR3的特异性扩增。Objective To study the CDR3 characteristics of T cell receptor carried by auto-reactive T cells from patients with rheumatoid arthritis. Methods BVI4 and BVI6 of the beta chain of TCR were amplified by realtime PCR, The length of CDR3 was identified by gene scanning. The sequences of CDR3 were analyzied by sequencing. Results The T cells infiltrated in the synovium of patients with rheumatoid arthritis showed oligo clonal expansion. The beta chain carried by infiltrated T cells was encoded by different VDJ gene fragments. The CDR3 had 15 bp, 21 bp and 24 bp length respectively. There were common motif among CDR3 area in infiltrated T cells. Conclusion The auto-reactive T cells infiltrated in the synovium are drived by specific antigens and therefore result restricted BV usage and common sequence motif among the T cells. The characteristics of the specific antigens that drive auto-reactive T cells need further investigation.
