摘  要:目的:观察幼年慢性癫痫模型大鼠情感行为及空间记忆改变,以及吡拉西坦的干预效应。方法:实验于2004-07/12在河北医科大学及河北师范大学生命科学学院完成。选用Wistar幼年大鼠50只。肌注马桑内脂注射液复制大鼠慢性癫痫大发作模型。每间隔3d重复肌注1次,经30次注射后,对其中点燃的动物改为14d肌注1次。造模3个月时点燃动物分为造模组、吡拉西坦低剂量组、吡拉西坦高剂量组、苯妥英钠加吡拉西坦组各10只。各组于造模3个月后开始连续灌胃,1次/d。吡拉西坦各组灌服吡拉西坦混悬液,分别为0.024mg/(g·d),0.048mg/(g·d);苯妥英钠加吡拉西坦组灌服苯妥英钠0.06mg/(g·d)及吡拉西坦混悬液0.048mg/(g·d)。用药1个月后进行相关指标检测。10只正常对照非造模组大鼠在造模开始时同期喂养并灌服等容积生理盐水。观察马桑内脂所致幼年大鼠慢性癫痫模型,达到点燃标准时动物情感行为的变化,以旷场活动性和拒俘反应性表示。观察空间学习和记忆能力的改变,以发现平台时间及搜索距离表示。结果:50只大鼠均纳入分析结果。情感行为观察结果:①旷场活动性比较:吡拉西坦组低、高剂量组以及苯妥英钠加吡拉西坦组明显高于造模组犤(5.2±1.2),(7.8±2.3),(8.4±2.6),(3.1±1.3)min,P<0.05或P<0.01犦。②拒俘反应性:吡拉西坦高剂量组以及苯妥英钠加吡拉西坦组明显低于造模组犤(2.07±0.38),(2.31±0.35),(3.91±0.98)min,P<0.05犦。空间学习记忆能力观察:①搜索时间比较:吡拉西坦高、低剂量组、苯妥英纳加吡拉西坦组在相应组次比造模组有不同程度的减少,而各个处理组内随训练次数增加平均搜索时间逐渐减少犤其中第1次分别为(45±9),(51±8),(44±9),(63±11)s;第3次分别为(42±7),(49±9),(42±7),(57±8)s;第6次分别为(34±8),(42±8),(35±6),(49±9)s,P<0.05或P<0.01犦。②搜索距离比较:吡拉西坦高、低剂量组、苯�AIM: To observe the changes of emotional behaviour and space memory in young rats with chronic epilepsy, and investigate the interventional effect of piracetam. METHODS: The experiment was carried out in the School of Life Science, Hebei Medical University and Hebei Normal University between July and December 2004. Fifty Wistar young rats were replicated into models of chronic major epilepsy by intramuscular injection of coriamyrtin, which was repeated every 3 days; After 30 injections, those kindled rats wee injected every 14 days. Three months after model establishment, the kindled ram were divided into 4 groups with 10 rats in each group: model group, piracetam low and high dosage groups and dilantin plus piracetam group, and the rats were began to treated with continuous gastric perfusion, once a day. Piracetam suspension was given to the rats in the piracetam groups (0.024 and 0.048 mg/g per day); Dilantin (0.06 mg/g per day) and piracetam (0.048 mg/g per day) suspension was given to those in the dilantin plus piracetam group. The related indexes were detected after 1 month. Ten normal rats were taken as controls, they were fed from the beginning of model establishment, and treated with gastric perfusion of saline of the same volume. The change of emotional behaviour when the coriamyrtin induced young rat models of chronic epilepsy reached the kindled standard was observed, and it was expressed with the activity in an open field and the resistance to capture. The changes of spatial learning and memory ability were observed, it was expressed with the time for detection of platform and searching distance. RESULTS: All the 50 rats were involved in the analysis of results. Observational results of emotional behaviour: ①Comparison of activity in an open field: It was obviously higher in the piracetam low and high dosage groups and dilantin plus piracetam group than in the model group [(5.2±1.2), (7.8±2.3), (8.4±2.6), (3.1±1.3) minutes, P 〈 0.05 or P 〈 0.01].
